Remimazolam Inhibits Neuronal Apoptosis in Rat Brain via Slit Guidance Ligand 2/Roundabout 4 Signaling Pathway

To investigate the effect of remimazolam on neuronal apoptosis in rat brain through slit guidance ligand 2/roundabout 4 signaling pathway. Primary cortical neurons were obtained from specific pathogen free 3 w old male Wistar rats and sub cultured cell culture dishes. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay detected viability; Hochest/propidium iodide double staining flow cytometry apoptosis. Western blot apoptosis-related gene expression pathway-related protein expression. In addition, after 2 inhibitor administration, changes levels key genes cells. Relative to control group, viability each group presented elevation depletion (p<0.05) a dose-dependent manner. With increase treatment time, Bcl-2- associated X protein, cleaved caspase-3 poly adenosine diphosphate-ribose polymerase showed gradual decrease anti-apoptotic B-cell lymphoma level relative 0 h (p<0.05). At same with time (0, 3, 6, 12 24 h), roundabout increased continuously h. Additionally, Bcl-2-associated up regulation down co-treatment group. Remimazolam pretreatment may inhibit 2/ pathway providing new ideas for clinical protection.
